The antioxidant system depends on production of NADPH for proper function. Its three major components in cells are the glutathione system, catalase, and superoxide dismutase [36]. The first is dependent on production of reduced glutathione by glutathione reductase that depends on NADPH. Catalase does not need NADPH to convert hydrogen peroxide to water but has an allosteric binding site for NADPH that maintains catalase in its active conformation. Superoxide dismutase does not use NADPH to convert superoxide to hydrogen peroxide but if this is not adequately reduced chemically by catalase or glutathione the increased hydrogen peroxide levels will quantitatively increase and inhibit the superoxide dismutase. Hence the entire antioxidant system depends on NADPH. As previously described, the need for NADPH from G6PD in embryonic stem cells with a complete knockout of G6PD made them highly sensitive to diamide (a potent oxidant) and led to more cell death than in cells with wild type G6PD activity [32]. This indicates that other cellular enzymes did not supply enough NADPH to protect the cells from cell death.

Need to concentrate on glutathione and catalase. I guess

Yes, but I guess there is need for Iron for catalase too right?
Thats why I thought of active B2. I think there could be a problem in UC for B2 to get active B2 (FMN or R5P)

B2 for glutathione reductase, active folate.

Do you think its possible to get iron on the transporters with active b2? In a pathogenic chronic disease?

And how do we incrfease NADPH ?
Problem with NADPH or upregulated NADPH oxidase?

Cysteine inhibits NADPH-Oxidase

NADPH will be increased only if potassium goes up and thyroid goes up. Since it is thiamine driven enzyme. manganese increases thiamine also which will increase NADPH also.. So zinc and manganese magnesium iron and b2 selenium iodine moly , could be an option to try. along with NAC and Nicotinamide

Glutathione redox needs FAD. and Fad will also depend on molyb and selenium. without them it wont be formed.

We need to try to speed up metabolism so G6PD starts working, this will increase glutathione redox. this will lower peroxide, which will allow SODs to work . once SODs are back online copper will be used.

DAO needs copper as a cofactor along with PQQ . may be lack of PQQ, is what causes methylation problems?

But how do we overcome low ceruloplasmin and iron bioavailability in general?

Taking iron supplements dont really go that well with UC. Free iron makes things really worse. Probably thats why there is so much copper instead of iron.

Looks like Sulfasalazine is bad for G6PD.

Nice article

Nice article

Ceruloplasmin goes up only in fast metabolism. You need to speed up metabolism. and feed the things at the same time. Since if you feed iron alone, your metabolism will go even lower. Since cortisol will go up. this is why people get constipation from iron pills.

to sum up, you think that UC is a problem with basically Glutathione recycling and other enzymes for ROS / hydrogen peroxide breakdown? And increase in ROS from heavy metals?

How do you think local mitochondrial fission chimes in there? Do you even agree on that?

Yes I tried/ am trying. But even if it helps, it will take time.

I dont really know where you see that his UC went away. He just has less toilet time with betaine :D
He had bleeding for ~1 year, I suggested him inuline and his bleeding went away after some days. Same with my last flares.

But this time its kind of different. It looks like more of a Histamine-driven Flare.
And Wuf does not have IBS. Or at least not that kind of "IBS" I have.

Of course there is some kinf of IBS while flaring in UC. But mine is worse when I'm in Remission than in flare.

I always thought of copper is used against high levels of Histamine in UC. And when the flare is over, upregulation of copper uptake is still there for some time and there is lots of copper. And iron gets displaced.
And with immunosuppressants, my body is probably slowed down (like a old person) and as soon as I get stress (even if it is really little), it seems like my metabolism speeds up and copper gets flushed out or used, I dont know.
I get adrenaline, sweating, dizzyness, and really nervous and wide awake & IBS and it feels like something toxic in my gut that wants out asap. But I'm not nervous or any kind of anxious before anything. It goes from 0-100 in seconds. And it does not differ in what I do.

For some weeks I tried caffeine 200mg to every meal and it really helped and all symptoms almost vanished. But at some point it stopped working as I thought I have burned adrenals.
As far as neurotransmitters,I tested them and all are low. But thats probably because just everything is low with immunosuppressants ?

Similar with smoking or Sports, if I speed up my metabolism slowly and am in this "increased" state, there is no IBS. But as soon I am "slow" and its a sudden increase, its IBS.
Im not sure if I can express myself right what I mean, tell me if I should try to explain it better. Its definately not IBS that is after some food intake or something. Its almost not there if Im at home or in literally 0 stress.
But with caffeine tablets for example, I could stand extreme stress with no IBS. Its like day and night. And its really not something in my head.
For ~30 min after Sport, I can do whatever I want and feel good.
Mornings used to be the worst. But now its similar the whole day long.

Pretty much everything that speeds up metabolism or brought me into flare, helped my IBS. Maybe because Copper could get used again.
And in the last flare ups, my IBS was almost gone.

In the past, the closer I got to flaring, the better IBS got.

But this time its different. Maybe I managed to get lower in Copper and all I need to do now is get in Remission and keep my copper-drinkink.-water faaaar away

When I was 15-18 I used to drink a lot of alcohol and other things and never had problems with IBS or UC whatsoever. Its weird that all started when I started to eat healthy and drink a lot of water.

Actually I dont really care about having UC. Just want to get rid of IBS and want to study and do normal things again. But Uc and IBS probably connected. But not the way that you need to get rid of UC to get rid of IBS. There is difference, as I said, in IBS from UC-flare and my IBS in remission.

I think you need to follow what I told you . You see everytime copper comes out with the bile, you have a leaky gut from IBS and copper and other metals enter the blood again free and cause problems.

May be you can try to use some kind of binders for the detox. like algin?

Everytime copper dump happens your potassium tanks. And that makes copper biounvailable. You need to support amino acids like wuf does, at least it will help with the detox.

RainCoat was in your situation with high copper. He followed copper detox , and as I remember UC and IBS went away
Just have to go with it. I guess. And keep your metabolism high .

Tbh I dont know what things wuf takes haha.

Wouldnt the best binder be CP ? But I guess hard to get in UC. ^^

So next thing I got hopes for is active b2. Maybe it helps me with copper, iron, ceruloplasmin...

So zinc manganese potassium magnesium iron b2 selenium iodine moly NAC Niacinamide. But will this help to get G6PD up?

Would spirulina or chlorella be ok insead of algin?

spirulina and chlorela I dont know. I know algin, since I took it. Spirulina and chlorela can cause the detox o their own. SO not sure if you need it. try to get some binder.

Do you have any idea how to get rid of high histamine levels? There are no enzyme-supps or things like that, that work on histamine levels right?
As far as I know DAO does only work on food as a supplement. Dont know about HNMT so far.
And antihistamines do only work on receptors.

@gbolduev Do you think metabolism crashes in UC but is kept upregulated local at inflammation in the gut?

I still don't know why you haven't just tried fasting in remission. You wouldn't have IBD during a fast, and the colon would have time to heal. It would reorganize the system so you could actually transport copper. Then refeed with good gut bacteria producing food to protect the gut.

Antihistamines don't just work on receptors when taken chronically. They lower HDC. That's why they don't work for acute treatment of chronic histaminergic diseases, but they do work when you take them for ~5 days.

Also things like G6PDH won't come online until ROS is under control. Ascorbic acid is damaged and mutated by ROS in IBD, and this disables G6PDH and pentose phosphate pathway, downregulating reducing equivalents, and creating a feedback cycle where ascorbic acid isn't properly regenerated because there's no reducing equivalents. The ROS is generated from metals, mostly from the breakdown of hemoglobin, or when you're in a flare it's from hyperactive mitochondria or increased inflammation, both creating superoxide.

The inflammation needs to be shut down, the body needs to catabolize defective tissues and re-appropriate metals back into healthy tissues or dump those metals in feces. Fasting will create a gut bacteria die-off, and you'll dump those bacteria along with metals in feces when you do have bowel movements - the rest will get taken in by whatever healthy tissue is left. There's too much iron floating around, the body can't control it - ferritin and transferrin can only go so high, and when they go high you get other problems as well.
